Happy to work anytime and overtime as required, but I need to mention that I have a prior commitment to the care of my daughters over the March and Easter long weekends. That being said, I am open to doing early shifts over theses weekends as the girls are late sleepers, so they won't require my supervision.

Ok, good. There is certainly nothing wrong with expressing your desire for notice. However, you may want to approach this, at least in the interview, by asking how the company assigns weekend or holiday work.
"Yes, I am very flexible and can make myself available on weekends and holidays. How is weekend or holiday work assigned? Is there a rotation? How much-advanced notice is provided?"
