It would not be in your favor to say that you are available for any shift, and then retract your availability because you aren't truly free once you get the job. Moreover, hiring managers generally understand scheduling conflicts such as having a family to look after are valid reasons for not being available to work at any time of the day. After advising that certain days or hours don't work for you, you can strengthen this answer by explaining that you are open and ready to work any night shift hours they may need you. By doing so, you demonstrate your willingness to prioritize your work.
