The hiring manager is gauging your response to determine if you are the right person for this job. What they really want to know is what differentiates you from other candidates they might be interviewing.
Read over the job description in detail and think of a few skills that make you uniquely qualified to perform the job duties. You need to be unique and separate yourself from the talent pool.
